The Q-NAb IgG Assay Principle
The Q-NAb IgG Test directly detects NAbs directed against RBM of SC2 Omicron BA.4/5 variant in serum or plasma by first blocking the binding of non-neutralizing anti-RBD antibodies (NNAb) from the sample using a novel human ACE2-BA.4/5 RBD fusion protein (FP). After blocking, the sample is incubated in a microtiter plate (MTP) well with recombinant SC2 BA.4/5 RBD protein immobilized to the solid phase. During this step, the human antibodies that are not blocked by FP and bind to the RBM on the solid phase are considered to be NAbs. Quantitation of the NAb level is accomplished by incubating the MTP well with a fluorescently labeled anti-human IgG antibody, thus generating a fluorescent signal that is directly proportional to the concentration of the NAbs in the sample. The fluorescent signal is read using an excitation wavelength of 560 nm and emission wavelength of 620 nm and NAb titer is calculated using a plate-specific calibration curve. This processed result is traceable WHO International Standard (1st International Standard for antibodies to SARS-CoV-2 variants of concern: NIBSC 21/338) and reported in IU/mL.
